I think the fight would go sometime like this: Sam shoots Duke in the face with the sniper rifle before Duke can even do anything other than maybe shoot Sam once or twice with either his shotgun or chaingun (which would barely hurt Sam), that would probably kill Duke considering how Sam's sniper rifle is 3 times more powerful than his rocket launcher, but if it doesn't it would leave Duke barely alive in which case he would either heal himself with a portable medkit in which case Sam would have enough time to shoot Duke with the sniper again, or Duke would use steroids and start running around shooting at Sam with the freezer which would probably never hit because of the low range, and Sam moving around too fast for any of those shots to land and Dukes other guns wouldn't kill Sam before Sam starts filling the room with cannon balls or riddles Duke with the minigun, or blows him up with the AS-24 devastator. Now given that from the start Sam knows everything about duke from what weapons he carries and how he fights from the start, and Sam having far more powerful weapons, he is at a very significant advantage from the start.
On top of knowing what weapons and equipment Duke has NETRICSA can also provide the most effective strategy to defeat Sam's opponents(assuming there is a way to defeat them). However none of Duke's tricks would work on Sam, because Sam would know to expect them, thanks to NETRICSA who can analyse any enemy Sam encounters, now i know the wiki says she only analyses enemies after Sam first kills them, but there are exceptions to that rule, at the end of first encounter, second encounter and SS3 she analyses the final bosses as soon as they show up, which leads me to believe that she can analyse enemies at any time they are encountered. Sam's weapons, in terms of damage, fire rate, and radius far surpass anything Duke has, but Duke's gear is much more tricky, he can set up traps with Lazer mines, he can make distractions with the holoduke, and he can move twice as fast because of steroids, which also triple his melee damage.
